In my experience, FlixBus buses tend to be a bit newer and cleaner than Greyhound. Plus, the WiFi usually works better. But honestly, for that long of a trip, both are gonna be a grind.

Most of the time a carry-on size bag and a personal item (like a backpack) are included. Just make sure the carry-on isn't huge, like airline overhead bin sized. If it's too big they'll make you check it.
